In addition to extensive applications in optochemical biology, we have developed genetic code expansion for the incorporation of crosslinking amino acids into proteins. Here, a diazirine is genetically encoded and used to map protein-protein interactions through photochemical activation and covalent bond formation with any biological molecule that is bound to the protein containing the diazirine. As a proof-of-principle, we have shown light-induced crosslinking of glutathione S-transferases in live cells.
